課題組介紹:張天真,求是特聘教授,博導,研究方向:作物遺傳育種。主要開展棉花結構基因組、重要性狀基因克隆與功能驗證、多倍體遺傳與分子進化和生物信息學等方面研究。
實驗目的:研磨棉花幼苗枝干提取DNA及RNA
實驗地點:浙江大學農業與生物技術學院
實驗材料和器具:
樣品:棉花幼苗枝干
儀器:全自動低溫冷凍研磨儀(上海凈信JXFSTPRP-CLN)、鋼罐一套
實驗步驟:
1、將棉花幼苗枝干剪成2-5mm長短的樣品;
2、將處理好的樣品加入鋼罐中;
3、加入配套的研磨珠子一顆,放入液氮種預冷幾分鐘;
4、將冷凍過的鋼罐樣品裝入凈信研磨儀中;
4、設定運行參數;
5、啟動儀器至研磨程序結束,可得到如圖所示樣品。

儀器介紹:
凈信上海凈信JXFSTPRP-CLN冷凍研磨儀是一種快速高效、多試管的一致系統。它能將任何來源(包括土壤、植物和動物的組織/器官、細菌、酵母、真菌、孢子、古生物標本等)的原始DNA、RNA和蛋白質進行提取和純化。廣泛應用于生命科學、醫藥、生化、環境、地質、農業、林業、質檢、材料等領域。

JXFSTPRP-CLN產品原理:
將樣品和研磨球放入研磨儀內(配置研磨罐或離心管/適配器),在高頻擺動作用下,研磨球在研磨儀內來回高速撞擊及摩擦,在非常短的時間內便可完成樣品的研磨、粉碎、混合及細胞破壁。
性能特點:
1.不破壞樣品成分:研磨溫度可自定義調節,能夠有效抑制核酸降解,保留蛋白質活性;
2.制冷可調:-50至37度
3.高通量處理量:一次處理多達192個樣品
4.操作簡便:內置程序控制器,可對參數進行設置;
5.安全性高:具備安全罩及安全鎖,無需涉及液氮操作,安全性高;
6.無交叉污染:在研磨過程中處于全封閉狀態,避免交叉污染;
7.低噪音:儀器運行過程中,噪音小于55dB,不會對其它實驗或儀器產生干擾。
8.無易損件設計,電磁鎖定保護,不銹鋼內腔,清潔消毒方便
9.重復性穩定:相同研磨程序獲得相同效果
