客戶介紹:浙江農(nóng)林大學(xué)位于杭州市臨安區(qū)、是浙江省重點建設(shè)高校、浙江省人民政府與國家林業(yè)和草原局共建高校。學(xué)校以農(nóng)林、生物、環(huán)境學(xué)科為特色,涵蓋八大學(xué)科門類的多科性大學(xué)。
實驗?zāi)康模?/strong>愈傷組織(callus)是指原植物體的局部受到創(chuàng)傷刺激后,在傷口表面新生的組織。它由活的薄壁細(xì)胞組成,可起源于植物體任何器官內(nèi)各種組織的活細(xì)胞。本次實驗對象為愈傷組織,要求磨細(xì),而且凍液氮磨,后續(xù)用來提RNA。
實驗步驟:
1、準(zhǔn)備JXFSTPRP-24L組織研磨儀一臺、離心管、氧化鋯研磨珠以及所需研磨愈傷樣品若干。
2、取適量愈傷組織、一顆氧化鋯研磨珠一起放入離心管中,蓋子蓋上。
3、將配備好的離心管放入液氮中預(yù)冷,凍脆樣品。金屬適配器底座也一起放入液氮預(yù)冷(預(yù)冷2-3分鐘)
4、將離心管適配器放入研磨儀中固定(適配器底部凹槽與軸的卡槽互相卡牢)放平后將離心管分別放入適配器對應(yīng)孔位,(注意平面配平,對稱放樣)蓋上適配器蓋,旋緊螺帽。
5、設(shè)置參數(shù)(根據(jù)所需研磨量微調(diào)時間),開始研磨。
6、磨好后打開儀器取出離心管,即可進(jìn)行后續(xù)實驗。

上圖為1分鐘的研磨效果,已經(jīng)完全研磨完成。效果非常好,老師要求愈傷研磨一定要凍液氮,否則rna可能降解,所以人工研磨特別不方便,經(jīng)過多次了解和對比所以老師們選用凈信研磨儀代替人工處理。效率高,效果好。完美解決了老師的研磨問題。
注意事項:
1、液氮操作需佩戴操作液氮的厚橡膠手套,注意安全。
2、放研磨管時時注意平面配平,只有一個樣品時需用空管配平。
3、凍液氮過程中一定是裝好樣品和珠子關(guān)蓋的研磨管,不要開蓋浸入液氮,液氮發(fā)熱膨脹容易爆管。
